3.2. Cav-1. peptide and Cav-1 knockdown promoted and inhibited astroglial differentiation and the expressions of NICD and Hes1 in NPCs, respectively
To verify the effect of Cav-1 in regulating the astroglial differentiation of NPCs, we artificially manipulated the level of Cav-1 and determined the expressions of S100β and GFAP in isolated cultured NPCs. By co-immunostaining of S100β or GFAP and DAPI, we found that the treatment of Cav-1 peptide, instead of control peptide, remarkably up-regulated the expressions of S100β and GFAP (Fig. 2). Immunoblot analysis further confirmed that Cav-1 peptide promoted the Dapagliflozin of GFAP, indicating that Cav-1 could promote astroglial formation of NPCs (Fig. 2). Meanwhile, we found that Cav-1 peptide, instead of control peptide, up-regulated the expressions of NICD and Hes1 (Fig. 3).
