SPC analysis. Cells were suspended in Cytofix/Cytoperm fixation/permeabilization solution (BD), and incubated with rabbit anti-human SPC UNC0321 (1:200 dilution) in blocking solution for 45 min on ice. Following incubation of cells with goat anti-rabbit IgG conjugated with FITC (Jackson ImmunoResearch, West Grove, PA) for 45 min on ice, cells were washed in Perm/Wash buffer, re-suspended in Perm/Wash buffer, and then analyzed by flow cytometry.
Statistical analysis. Statistical significance was determined by paired t-test analysis. Data are reported as mean ± standard error of the mean (SEM). P cells for each marker is shown in Fig. 1C (data for HS68 fibroblasts are not shown).
