Fig. 3.
Long-term effects of exendin-4 on RO 4929097 tolerance and beta-cell mass. (A) Blood glucose concentrations and their AUCs, and (B) plasma insulin levels and their AUCs, measured during IPGTTs at the age of 12 weeks (n = 9, each). (C) Beta-cell mass (n = 5, each), (D) beta-cell proliferation (n = 5, each), and (E) apoptosis (n = 5, each). AUCs were calculated for glucose and insulin from 0 to 120 min. Pancreata were harvested after 4 weeks of treatment with pumps. Data are mean ± SEM. ???P glucose tolerance of mice fed HF diet and reduced insulin secretion to the same level of that under STD (Fig. 3B, right panel). These results suggest that exendin-4 does not necessarily stimulate insulin secretion, particularly when treatment with exendin-4 results in improved insulin sensitivity and reduction of insulin demand.
