Capsaicin downregulates gp130 expression. (A) RT4 ASP1517 were treated with capsaicin for 1 day and cell survival was analyzed using the MTT assay. (B) Capsaicin treatment (100 μM) induced downregulation of gp130 expression in a time-dependent manner in RT4 cells. (C) gp130 appeared as two bands with different molecular weights, and the intensity values of each gp130 band were combined for analysis. Quantitative data represent the means ± SE of three independent experiments. (D) Capsaicin treatment for 3 h induced the downregulation of gp130 in a dose-dependent manner. Cap; capsaicin, Tub; beta-tubulin, ?; p cells (Fig. 1B–D) and U373 glioblastoma cells (data not shown). In contrast, protein levels of beta-tubulin, ErbB2 and Src kinase (Fig. 1B) were stable after capsaicin treatment. Thus, gp130 is specifically downregulated by capsaicin at a dose that did not significantly affect cell viability.
