We recently reported that gp130 is subjected to endocytotic degradation after treatment with reactive aldehydes in neuroglial tumor ABT-538 [16]. In this study, we examined the effect of capsaicin on gp130 stability in neuroglial tumor cells.
Materials and methods
Cell culture. Rat schwannoma RT4 cells and U373 glioma cells were obtained from the American Type Culture Collection and maintained as previously described [16].
Survival assay. The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was performed to analyze cell survival as previously described [16]. Briefly, cells were plated in Dulbecco’s modified Eagle’s medium (DMEM) containing 10% fetal bovine serum (FBS), and 1 day after plating, cells were washed twice with PBS and grown in serum-free DMEM for 1 day in the presence or absence of the indicated reagents, followed by incubation for 1 h in the presence of 0.5 mg/mL MTT (Sigma). The medium was then aspirated, and the cells were dissolved in isopropanol (100 μL/well). Measurement of the reaction products was performed in a microplate reader (Bio-Rad). All experiments were repeated a minimum of three times.
