Such low level of genetic differentiation
To assess the effects of nitrated fatty acids on the regulation of cell adhesion molecule expression, we examined mRNA and protein levels of selected adhesion molecules in TNFα-stimulated HUVECs pretreated with OA-NO2 or LNO2. Both OA-NO2 and LNO2 significantly inhibited TNFα-induced CCT129202 of E-selectin, ICAM-1, VCAM-1, and MCP-1 mRNA (Fig. 3A). This suppressive effect was reflected on the protein level (Fig. 3B–E). Based on a densitometric analysis of five independent Western blots, pretreatment with OA-NO2 suppressed the expression of TNFα-stimulated E-selectin (Fig. 3C), ICAM (Fig. 3D), and VCAM-1 (Fig. 3E) by about 50%. LNO2 also attenuated TNFα-stimulated adhesion protein expression, but to a somewhat lesser extent, similar to the differential effects of these agents on cytokine production. Consistent with previous reports by Wright et al. [20] and Ferreira et al. [8], OA-NO2 and LNO2 also induced HO-1 expression at the mRNA and protein levels (Fig. 3). The nitrated fatty acids did not affect PPARγ expression, and neither of the corresponding native fatty acids affected adhesion molecule mRNA or protein [removed](Fig. 3).
