Expression pattern analysis of the putative
Genetic requirement for Notch signalling to initiate NFIA MC1568 in the spinal cord. In PS-1?/? embryos Hes1 expression was strongly down-regulated (A and B) while Hes5 expression was unaffected (C and D). NFIA expression was almost abolished (E and F) while Glast expression was strongly reduced except in a small region in the vSC (G and H). Fgfr3 expression at e13.5 was almost abolished while OLIG2 was expressed in a somewhat enlarged pMN domain.

Supplementary Fig. 9.
Genetic model interlinking different genetic pathways upstream and downstream of SOX1 during gliogenesis. The core components of this model can be summarised as follows: (1) Notch signalling induces a pro-astrocytic program of glial specification in the pMN by inducing the expression of NFIA in the VZ, in PAX6/SOX1/HES1/HES5-independent manner. (2) PAX6 and Notch signalling positively regulate independent mechanisms that affect SOX1/HES1 expression that inhibit MASH1 expression and thus MASH1-dependent OLP specification. Green arrows indicate induction; red arrows indicate repression of expression or of a mechanism. OL, Oligodendrocytes; AS, Astrocytes.
