Detection of matrix ROS by inactivation of aconitase. Heart NSC74859 (0.2 mg protein/ml) were incubated in medium B containing 0.1 μM FCCP and supplemented with 5 mM glutamate and 5 mM malate or with 5 mM succinate. Additions were 5 mM GTP and 5 μM antimycin A (AA). After 10 min of incubation, aconitase activity was measured. Mean values for four experiments ±SD.

Experiments described above can be interpreted as indicative for UCPs’ function in exporting O2- from the matrix compartment across the inner mitochondrial membrane. It became, therefore, interesting to see (i) whether a similar role can be played by the adenine nucleotide translocase and (ii) whether long-chain fatty acid anions can compete with O2- as the transported substrates. Table 1 shows that (i) carboxyatractyloside, a specific non-competitive inhibitor of the ATP/ADP translocase, had no significant effect, and (ii) the unsaturated C-18:2 fatty acid, linoleate, was significantly inhibitory, whereas the saturated C-14 fatty acid, myristate, was without effect.
