3.4. siRNA against Twist inhibits hypoxia-induced VM
To further assess the possible correlation of Twist Adrenorphin with hypoxia-induced responses, the effects of specific Twist siRNA on VM formation were evaluated. We used three siRNA duplexes specific for human Twist and scrambled double-stranded RNA unrelated to any known gene as a control. The results showed that Twist-siRNA-1 significantly suppressed the expression of Twist (Supplementary Fig. 1). It was the most effective RNA interference sequence. In the following Twist siRNA experiments, Twist-siRNA-1 was used to examine the effects of hypoxia on Twist-depleted cells. Based on the changes in cell VM networks (Fig. 4A), we concluded that Twist suppression blocked hypoxia-induced VM, whereas the double-stranded RNA control showed no effect on VM networks (P expression ( Fig. 4B).
