Love Actually is a classic. They've tried to make similar movies since it came out, but none are as good as the original.

I was also going to suggest Love Actually. It's not my favorite, but i like it. Personally one of my favorites is About Time, coincidentally, also by the same direct of Love Actually. I turned out to like About Time more than i had anticipated. It wasn't what i was expecting. I'd also like suggest The Holiday.
I think it ultimately comes down to your kind of comedy. Are you a Mama Mia kind of person, or Knocked Up, or Shallow Hal. What kind of humor do you like? are you looking for something with a mix of drama?

The 40 Year Old Virgin. xd Although I suppose that leans more towards comedy than romance.
It states that it's a romantic comedy, but 500 Days of Summer is really good as well.
