Is there any way to get Shadowmere without joining the Dark Brotherhood? And does anyone in the game ever give you a quest to annihilate the Dark Brotherhood?

As far as I know you can't get Shadowmere any other way then completing Dark Brotherhood quests. As for annihilating the Dark Brotherhood, I'm not sure if you can except for when you get taken to do the initiation and just kill her right away.

No. Unless you use cheats (if you're using the PC version), you can't get Shadowmere without being in the Dark Brotherhood. You could get Frostmere, however.

To get the quest Annihilate the Dark Brotherhood, you have to kill Grelod the Kind, like you were planning on joining the Dark Brotherhood. After getting the note with the black handprint, go to sleep, and wake up in the cabin. Instead of killing the hostages, attack and kill Astrid. Take the key from her and leave the cabin. Find any guard in the city and report to them. They'll tell you what to do from there.
