Are they the only thing that can kill a Timelord? I know that if a Timelord chooses not to regenerate, they die, but can the blast of a Dalek surpass that and actually kill a Timelord? Also, does The Doctor lose some of his lifeforce with every regeneration?

The Doctor got shot by a Dalek at the end of The Stolen Earth and managed to pseudo-regenerate, so apparently the Dalek death ray isn't enough to outright kill a Timelord.
It used to be that Timelords were limited to 12 regenerations, after which they permanently die, but it's unclear if this restriction still applies to the Doctor. If it does, then he better take out a life insurance policy, because he doesn't have very many regenerations left.

of course, unless the show takes a massive ratings dive, Steven Moffat will come up with a way to surpass that restriction.

Either Steven Moffat or the next head writer that comes along will find a loop hole. I mean, RTD came up with the Time War.

Well, according to Turn Left, if the doctor is injured without enough time to regenerate he can die.

Frantic Femme Fatale Well, according to Turn Left, if the doctor is injured without enough time to regenerate he can die. That seems to be the primary loophole for now. Ten's just lucky he had that spare hand lying around (in a jar) or he'd probably have regenerated already.

I wonder if the Doctor could regenerate if he was decapitated and had a wooden stake driven through both of his hearts.

Something tells me that the brain is required, and if he were decapitated, he would just die. I also think that he would die if both of his hearts were damaged, as we saw that he was in a lot of pain when one of his hearts went dead in Shakespearean Code.
