I think it depends from person to person. I personally think it's Rei in the anime and manga, though they fight and whatnot alot.
(BTW, I would say Molly in the anime, but we don't see hardly any of her after Serena becomes Sailor Moon, or rather, we don't see her as much as the first season progresses)

i'd say that all the inner scouts are her best friends. she hangs with them all the time and they have been with her for a long time.

Again...it all depends on what your watching...
in PGSM its definatly Ami...
In the anime its Rei
I havent read enough of the manga to base a decision
and in Sera Myu it is Ami...

In the anime, I'd say it's definitely Rei.
They fight a lot, but whenever anything happens to Usagi, Rei's always the one comforting her. Also, in any of the scenes when the characters die, the animators and Usagi focus on Rei a lot more than the other senshi. That's always kind of annoyed me a little, actually.

and the manga its..minako..xD...but i think its more like rei..she always get mad at usagi xD..thats so funny

Rei is the most common answer and I find it to be true since she's the one who cares the most about her but in sera myu I would think that her friendship is shared but she does lean a little to wards Ami though.
Just my opinion though.~

Well, I guess I have a hard time seeing Rei as the best friend because especially in the first season Rei does a lot of things just to try to spite Serena. I definatly think Rei cares about Serena but I think that Ami or Lita are overall more supportive of Serena consistantly through out all the seasons.
So I guess if we take Molly out of the running, which I guess is kinda true that she basically fades away after season 1, I would say Ami or Lita.
Now in the R movie, it's definitely Rei. I mean when Sailor Moon dies everyone is putting on a brave front except Rei who is balling and basically throws herself on top of Serena. In that one I definitely see her as the best friend.
