I'd want the thing to have hybrid nuclear electric thermal propulsion, Antimatter propulsion, or nuclear photonic propulsion. They're kind beyond current technology though. Hybrid nuclear electric thermal propulsion is theoretically possible with current technology, so let's go with that. Let's make it a big, orbital ship (probably bigger than anything we actually have) Transportation to and from the ship is done by some sort of landing module. The ship is big enough for necessary supplies. Maybe not economically logical though, as I said before....
