It takes place between 3 and 1... so no.
Its basically another prequel.

DMC 3 took place before 1, remember?
So DMC4 picks up some years after 3, but before Dante gets hired by Trish to go to Mundus' Island in DMC 1.
Nero is some dude protector in an Order of Religion that worships Sparda. According to the story so far... Dante shows up in the city and starts killing all the members of this Religion/Cult, and Nero has to stop him.
So the main character is Nero.
Iunno.
During all this, Vergil is still in Hell getting brainwashed and trained for his appearence in DMC 1.
