I believe Ian, Gambino, and I believe that now Edmund are all using this serum. They've all been shot (according to the comic, Edmund knows of the Sniper and has encountered him before), so they should all be dead. However, each one of them has used the serum to save themselves in different ways. This serum gave Gino super-powers, yes, but it also did something else, namely, it made Zombies at Halloween '05. Are you thinking what I'm thinking? This serum allows them to stay alive and not be zombies. The Von Helsons have been revived as Vampires to try and kill the Gambino family and all those involved with them. If you talk to them, their comments make them sound like vampires. Trust me...

Actually, I don't think they meant anything by "They knew that bullet wouldn't kill Ian". They were talking about how 'they' were just trying to scare Gambino, Edmund and Ian in order to get a secret. I think they were just saying "They could kill us easily if they wanted, but they failed their shots on purpose", (and that's why) [they knew that bullet wouldn't kill Ian].
