This is what I KNOW:
Two fan favorites die.
It'll be in the same episode that Sam and Dean fight Lucifer.
Episode ten has a strange title "Abandon All Hope" and is the final episode of 2009.
It's going to be epic
Jim Beaver either has a horrible sense of humor and was trying to be funny, or he was being serious in a joking manner: as in, party episode ten like it was the last time.
Episode ten, Castiel's story arc is going to start in episode 10 - where Lucy captures him in a ring of fire and interrogates him.
Ellen and Jo ARE coming back in episode ten.
All five are going to be in that episode.
Anna IS coming back.
Jimmy's death is not currently canon.
Random fact: Kripke thinks of Sam and Dean kinda like Han Solo and Luke Skywalker. Food for thought. razz
Kripke quite clearly does not want to water down and has basically every intention of ending it with a bang this season. (This MIGHT not happen, but we do KNOW that he wants both those things to an extent)

NOW. IT'S TIME FOR ME TO GO OVER THIS...again, and think about everyone elses responses so far. razz
First of all: If Bobby doesn't get his damn legs back tonight, I'm calling shenanigans and voting him off the island.
Love him. Really. Is he in the way? Yes. When heroes get to the climax of their tales, where do their mentors go? Six feet under. It's part of the hero cycle. Obi Wan Kenobi, for example. Or in more extreme cases, like, say, Harry Potter: Lily and James, Sirius, Dumbledore...(any adult figure with more knowledge that could help in his path to growth and defeating evil and whatnot).
Basically, in the hero cycle, which Sam and Dean clearly fit into - you end up killing off the help. The hero ends up fighting it's antagonists alone. You have the friends, you know, the comrades, that fight the battle with the hero, but they tend to play their role out differently. A lot of times, they live. Ron, Hermione, Princess Leia, R2D2 - WHATEVER. YOU GET IT.
As I see it: Sam and Dean are Han Solo and Luke - or both Harry Potter. Cas is Ron or Leia (...xDDD Okay. Brain went THAT-A-WAY, ignore this), or whoever. It's not about him, but he is in a similar boat. He's not a MENTOR, as he's just as lost, but he is good help.
Like we've all said - he's basically a Winchester. But not a John Winchester - more like another Sam. razz He's in the hero cycle loop. He COULD die, but I have a few issues with it that I'll address in ONE. MOMENT.
First I have to bring this up - from what we know, Episode Ten involves a reason to go beat the hell out of Lucy - what? Don't kidnap Cas, ********. D: We also know that episode ten has a 90% chance of being the episode in which fan favs die as we know it's epic, involves Lucy, and has a cliff hanger (oh, did I forget to mention that one? razz ). What better way to hang us by killing people off.
I believe we probably won't learn if one of the two actually died until Supernatural returns in January. We also don't know for sure that they won't PRETEND to kill more than that.
So where I'm at now - I think this spoiler is talking about episode ten. And I think Bobby is one of the two, or at least a pretend death. ;D HOWEVER, he's a mentor, and I'm surprised he's lasted this long. And now he's essentially useless (depending on tonight's episode).
Now for my Castiel reasons:
1) He died already. I think someone asked at a con that I was watching, if they were killing him - and one of them wondered aloud "Hadn't we already? razz " - I think, to be honest, it'd be silly to kill him again. Main character initiation, guys. At most he'll die in the finale, but I think they look at him like we do - one of the boys - who clearly won't die, unless it's the last episode. I strongly believe we have Castiel for keeps.
2) If I'm right about episode ten being the episode with the deaths, Misha's contract is a dirty rotten liar, and I need to have a talk with it. Also, they've already started spreading out his five episodes that he can't be in.

I hear he's in Changing Channels, but that was an iffy source, so I won't continue my list right now.
3) If Princess Leia (or Ron) gets to live, Castiel is damn well just as important and worth keeping alive.
4) It's serve no other purpose than to make me cry - and I highly doubt Kripke is out for my tears in Cas's case. He upgraded him to main character for us, why would he bring him back just to slap our Misha-lovin' faces? It doesn't affect his story telling, and he likes to keep us happy - I just can't see Cas dying (not in episode ten for SURE, and not in the end, either.)
As for Chuck? He was the first person I thought of alongside Bobby. It'd be like weakening the heroes. As was said, he has an epic power, and that's no good. He is clearly dispensable in that he's too much help and not developed in any worthwhile way. We had Ash, who was super useul - Kripke killed him. He got in the way of the hero cycle. Chuck is becoming less useful, just like Bobby, and they've served their purposes well. I love them, but I'm voting them off the island.
As for Anna - I like to hope the b***h is already dead.
ELLEN - there are casualties in war - not to ruin the end of HP, but everyone and their cousin's maid was a casualty. razz I don't think she's going to be in the EPIC TWO PEOPLE DIE AND WE FIGHT LUCY thing, though. They won't make that about her. If anything, she'll die a little later. I love her, too, but really.
I think Kripke or some creator said this: It's basically four good guys against the world.
...Dean, 1. Sam, 2. Castiel, 3. Bobby, 4.
Bobby, as I've said a lot now, is the mentor, Castiel's the friend. I'd kill Bobby before Cas (and not because of my love exactly, but storytelling-wise, that's the best option.)

What I don't know is if Chuck will be in the episode. Or if we'll see all the deaths onscreen in the episode that it happens.
