Effect of TNF-α on AQP3 mRNA expression in DJM-1 cells
Next we examined the effect of TNF-α on AQP3 mRNA expression in DJM-1 cells. TNF-α significantly decreased the level of AQP3 mRNA. The effect of TNF-α was dependent on treatment time (Fig. 2A) and concentration (Fig. 2B). The effect of TNF-α concentration on mRNA levels was consistent with the decrease in AQP3 protein expression as AQP3 mRNA expression in cells treated with TNF-α (20 ng/ml) for 24 h was about 40% of that in control cells. As shown in Fig. 2C, treatment with TNFR1 small interfering RNAs significantly inhibited the decrease in AQP3 mRNA expression caused by TNF-α. These data suggest that the TNF-α-induced decrease in AQP3 protein levels in DJM-1 cells is due to a decrease in AQP3 mRNA expression.
