The calvaria of E13.5 mice were dissected and cultured in a Trowell-type culture dish. The Foxc2 construct was transfected into suture mesenchymal Olmesartan by electroporation whereas Foxc2-siRNA or control-siRNA was transfected using the transfection agents described in the Materials and methods section. After 48 h of culture, the calvaria was harvested and fixed. Positive brown-stained cells for BSP were indicated by the arrows in (H). HE, ALP, and BSP staining were performed. All scale bars represent 100 μm, except for G and H, which represent 25 μm.

After 48 h of Foxc2-siRNA transfection, ALP staining was rarely observed in suture mesenchymal cells and adjacent parietal bone of the calvaria (Fig. 2L). However, the control calvaria showed ALP staining in the parietal bone as indicated by the arrow (Fig. 2K). Thinning of the epidermis and dermis was also observed as well as reduction in the number of hair follicles in Foxc2-siRNA treated calvarias (Fig. 2I and J).
