If you read the manga, it's revealed in the VERY FIRST issue: in the first two 'acts' no less. Kaoru suspects he's 30, but he doesn't know and he looks younger than that (hsc-cha! I look older!) so they settle on 28 and be done with it.

He's been 28 since about the mid 1990's. I hope it stays that way... And what Silver Bells says is true... I think they even announce it in the first or second episode... I LOVE that anime... heart

Going, going...
In the manga somewhere there was a page that revealed all the ages of the current characters of that volume, and it had Kenshin down as 28. ^_^x
