Usually, most of these sign-up spots ask for a few days advance warning, since it takes time to gather crew mates for a tuna and get funds for the power-ups used during the run.
For the run, you'll need at least one pack of Revives, one pack of Ghi Amps (Regular or Deluxe), and 3-6 hours of spare time to dedicate to your computer.
What Hildr said is the usual DPS Ringset... 2 Buffs | 4 Attacks | Passive | [Optional Slot: Meat, Attack #5, Passive #2, or Adren]
Having the scaling buffs at 12.0 makes it a lot easier for your crew to assign, but most crews don't expect their tunees to have leveled buffs, so don't worry too much about that.
