Typically the most effective way is to have an amalgam of defining characteristics you feel confident you can reproduce and practicing the living bejeesus out of them. There's of course the defining characteristics that stand out immediately -- birthmarks, scars, piercings, tattoos, hairstyles, clothing, signature accessories, etc. -- but it's dangerous to rely on those. Some things to think about: overall face shape (heart-shaped? square? round? etc.), how much you define the cheekbone/how high it is on the face, eye shape (particularly important when it comes to manga, where there's such heavy emphasis/a lot of attention on the eyes), eyebrow thickness, and general body shapes (waist-to-hip ratio, leg length, shoulder breadth, GIANT BAZONGAS vs. itty bitty titties vs. everything in between, etc.).
