Depending on how far along you are and which game you're playing, you may have noticed that after you get the National Pokedex, a new path opens up along Route 214. This path leads to the fourth large lake, Sendoff Spring. (Note: In Platinum, you arrive here after battling Giratina. In Diamond and Pearl, you'll have to make the trip there yourself.)
There's not anything super-special about Sendoff Spring itself. There are no tiems to collect (except possibly with the Poke Radar), and all of the pokemon here can be caught elsewhere in the game (with the possible exception of Chimecho). You will need Rock Climb to get the cavern, though, and once inside the cave, you'll need Rock Smash and Defog.
Turnback Cave itslef can be very confusing if you don't know what's going on. There are basically three main rooms, or Pillar Rooms, that have a large stone in the center with writting on it. When you read the text, it will tell you two numbers: the top number is the number of Pillars you've seen and the bottom number is how many rooms you've been through.
To get to Giratina, you have to find the third pillar room. But there are a few catches. The 18 other rooms in the Cave appear completely at RANDOM, so there's no direct path to Giratina. Furthermore, if you can't reach Giratina within 30 rooms, then the next room you enter will be the starting room. Also, if you go back through the same door you entered from, it will take you back to the first room.
So basically, getting to Giratina basically amounts to luck and persistence!
The pokemon inside include Bronzong, Bronzor, Golbat, and Haunter. Fire type moves and dark type moves should work effectively on all of them. There are no items in here initially, but check back after you catch Giratina to play a sort of mini-game.
When you finally get to Giratina, you'll find he's at level 70 and knows Shadow Force (ghost), Heal Block (psychic), Earth Power (ground), and Slash (normal). Heal Block probably won't be much of a problem, but Slash has a high critical hit ratio which can be bothersome. Earth Power can be dodged by pokemon that know moves like levitate, but Shadow Force can hit even if you use moves like Protect. Shadow Force is also a two part move: Giratina disappears on the first turn and then attacks on the second.
Since Giratina is part ghost type, he can be tricky to catch. Normal type moves won't work on him, but conversely, his Shadow Force move won't work on a normal type pokemon. If you have a normal type with a couple of special attacks, it might fare better against him than some others. Moves like Aerial Ace, Swift, and Magical Leaf can also be helpful since they're unavoidable, so they should hit Giratina even when he disappears. Ice, Dragon, Ghost, and Dark type moves will be super effective as well.
As far as which balls to use, I'd suggest Dark Balls to start, then Ultra Balls and Timer Balls if the battle runs long.
After you catch Giratina, you should try returning to Turnback Cave to play a mini-game of sorts. This time, when you find the third pillar, you'll find a reward based on how many rooms it took for you to reach the third pillar. The rewards are as follows:
3 rooms - Reaper Cloth 4-15 rooms - Rare Bone 16-30 rooms - Stardust
The last two can be sold to shops at a high price, while the Reaper Cloth can be used to evolve Dusclops into Dusknoir when its traded. If you can't get the Reaper Cloth here, you can also find one at Route 229.

sounds good. and as far as the cloth it can also be used as a bargining chip with friends who need it just FYI

No Giratina wiped out my whole team I was grossly unprepared for it as was busy trying to hatch eggs. I just had two of my main Pokemon with me. crying I haven't found it anymore. I was a little mad too since I had to just quit without saving cause I had just hatched hoot-hoot.

moves like -ace Magical leaf and Swift wont hit him when he disappears
those mvoes work with where the pokemon was "standing" before using moves like dig, dive, bounce, fly, or Shadow Force
or rather the sprite must be seeable to land those hits and if it's there i'ts a hit except for swift on ghosts
