My uni portfolio contained... life drawing, a variety of textiles (because that was what I was doing before illustration), mixed media, digital work, typography... I'd take a sketchbook, they're invaluable for looking at your working process xx

What kind of course is it, and what do they expect you to know before going in?
I would second what miserabelle recommended, along with perspective work and/or environmental sketches, photo-realism or photo-reproduction (one of each would be ideal), a painting or two in traditional mediums. I feel like I'm forgetting something...yup, big help I am.
But Life Drawing is probably a definite. Can't go wrong there. If you're not 100% certain of a piece, maybe leave it out, or get someone you know in the same course or a similar program to get an opinion of it. One of my instructors would say something like an eight piece portfolio of 'A' work stands better than a 12 piece portfolio that's two thirds 'A' work.

I think something to think about is the order that you display your work in your portfolio. A lot of people will put their strongest work first but I think that can leave the rest of your portfolio feeling a little flat and empty. I'd spread out key pieces throughout to keep your interviewer's attention. I think it helps make your portfolio more solid xx

From a salesmanship point of view, expect most people to make up their mind early, so if you have them sold by the time they get halfway through the portfolio, then you don't have to change a potentially negative opinion in the second half. First impressions and all that...
