Is the 14th related to Mana? I mean, if not, why would the Earl kill Mana? I'm a bit confused here. ><" Not to mention that Mana taught Allen the song.

Mana and the 14th were brothers. The Earl didn't kill Mana, Mana just died of an unknown cause. Allen then brought him back as an Akuma, which is when Mana cursed him (hence Allen's eye), then Allen's Innocence activated and killed the Akuma version of Mana.
