As far as I'm aware, NO ONE is canon for Snape's sister, so I'll have to guess based mainly on your other clues.
Minerva McGonagall?
(If that's who you're referring to, yikes. Someone didn't do their math. She's in her late seventies, and Snape's in his early thirties. Honestly I think Irma Pince is related to Snape, but not as a sister, and she doesn't fit the clues since she's not actually a teacher, and certainly not a head of house.)
1) Muggle Studies Professor
2) Captured and tortured and killed by the Death Eaters and Voldemort.
3) Draco couldn't keep his eyes off of her as she hung in between tortures.
