か is not usually use with colloquial speech to make question. It often use to consider whether something true or not or to be making suggestion.

の is question in colloquial.

Changing ない to れん

Ex: Does 読まれん work for 'can't read?'

Also when one adds 「って」 to something that isn't a verb, what is it's significance/is it actually colloquial, or rather just changing the tone of how your say it?

Ex: がんばってツて I have read before, but I didn't get what it means.

the anser to your first question is that it it s a contraction of the negative passive (れない=れん)

it is the thete form of ganbaru- ganbatte

it was probably a little tsu you say and te would be the te for of the the colloquial ~teite- ~tete (te form of teiru)

So the second "te" is said in regular speaking Japanese?
