@amakama: No, she means if like you were the director, and you had to pick actors to play the characters different than the ones already there.
Anyhoo. I personally wouldn't change a thing. The choice in actors in POTC is perfect, and one of the reasons why it's so exemplary.

Amen. But, since you asked the question....
Elizabeth - Natalie Portman (Knightley's cute, but Portman's deeper)
Will - Toby Maguire (Bloom may have the hotness factor, but nothing says "emo" like Maguire. Besides, I bet it'd be funny to hear his British accent)
Jack - Johnny Depp (seriously. He's the ONLY one that cannot be replaced. The RockStar Pirate cannot be played by anyone else. If it HAS to be someone else, it would have to be...NO ONE!)
Davvy Jones - Hugo Weaving (that's right. Agent Smith, baby. Do you fear death, Mr. Anderson?!?!?)
Barbossa - Bill Nighy (In all honesty, though, no one can laugh more maniacally than Geoffrey Rush. Hey, Kevin Spacey came in at a close second here...y'all got lucky!)
Tia Dalma - Jada Pinkett Smith (and not only would I change the role, I'd change the makeup to match it! God her teeth were horrible!!)
....ya, chances are y'all might pelt rocks at me....or worse....but I just think it's funny...haha!

Will- Ohmehgosh... Owen Wilson! Awesomeness! (A young Billy Crystal or Tobey Maguire would be pretty funny though.)
Jack- Unchanged
Davvy Jones and Barbosa- Gerard Butler could play both of those roles well.
Tia Dalma- Not important
