It's definitely rough right now, but I was wondering if I could get some opinions on how to really get some nice color and definition going.
Any and all help is really happily accepted.

I would start blocking color into the background, keeping lighting in mind. I find that if the background isn't done early on, the subject matter tends to look copy and pasted into the piece. I would also wait to start defining details until colors are blocked in.
I'd have started with the bg first. But shadows and highlights can be put in later. Thankfully, digital work isn't so hard to change later.
