Battle Arena: A massive abandoned church, rows and rows of pews and marble pillars that extend to the ceiling. Two stories tall, eight sets of stairs throughout the church allowing access to balconies above.
Contestant 1: Spike Spiegel. He has his Jericho 941 pistol, with extra ammunition. He also has grenades and remote control explosives. Keep in mind his skill in Jeet Kune Do.
Contestant 2: Kawamaki Gensai, the real life inspiration for Kenshin Himura. Most lethal of the four hitokiri of the Bakumatsu period. Fighting style is the 'lightning fast' Furanui Kenjutsu, otherwise known as Shiranui-ryu. Unlike his animated counterpart Kenshin, his speed is not inhuman but very impressive. He wields a traditional katana and wakizashi.
Contestant 3: Jigen, marksman ally of one Lupin the third. Jigen can draw and fire his gun, with amazing accuracy, within 0.3 seconds. He is armed with two pistols and enough ammunition to reload throughout the fight.
Contestant 4: Captain Kuro. Able to move at high speeds, to the point where he is practically invisible, although in this state he can't see what he's attacking and just slashes at everything in his path. Wears two pairs of gloves with a katana extending from each fingertip, to replicate claws.
Contestant 5: Lloyd Iriving, wielding two katana. For those familiar with Tales of Symphonia, this is before his angelic transformation and also before he gets any of the really spiffy blades near the end of the game. Focuses on above average speed and powerful strikes, including one attack where he leaps into the air and then slams down towards his opponent, generally tearing through them.

I would say Kuro.
He easily beats Gensai, because Gensai, for all his skill, is only human and doesn't have superhuman anime prowess.
Spike would be a little tougher, but still Spike is more of a realistic character, and would get beaten because of that.
Jigen is very fast and accurate with his gun, but Kuro can just move so fast Jigen wouldn't be able to target him.
I don't know anything about Irving, so I won't comment on him.

I agree with Das. Minus the Symphonia guy who I know absolutely squat about, Kuro can easily beat the other three.

I'm actually going to give Spike the win.
I'm figuring that Jigen would take down Gensai, since he can draw and fire his gun much faster than the Gensai can close the distance between them and kill him. Meanwhile, Spike makes his way up to the balcony, placing a bomb near the stairs as he does, and picks off Lloyd from above.
Kuro tears Jigen to pieces. Jigen has great aim, but Kuro is just so fast. Assuming he isn't moving at his highest speed, he rushes towards the stairs to attack Spike, planning to just deflect any incoming bullets with his 'claws.' Spike detonates the bomb he's placed on the stairs and blows Kubo away.
If Kubo can leap back to dodge the bomb's explosion, Spike shoots him while he's in midair. Speed means nothing in midair if you can't fly.

Going with your "shooting him in midair" strategy, what prevents Kuro from blocking Spike's bullets in mid air?

Surprise. Kuro just evaded an explosion.
Of course, another factor I initially failed to take into account is all of the smoke and debris as a result of the explosion. . .on the one hand that means Kuro wouldn't see the bullet coming in time.
On the other, it means Spike wouldn't be able to aim properly.
