Determination of infarct volume and Cyproheptadine hydrochloride swelling. Five rats each from the GSK-3 inhibitor VIII and saline groups were sacrificed by intraperitoneal injection of sodium pentobarbital (150 mg/kg) at 2, 8, 24, and 72 h after occlusion; five rats from the sham group were sacrificed 24 h after the operation. Infarct volume was calculated using the same 2% 2,3,5-triphenyltetrazolium chloride (TTC) (Sigma, St. Louis, MO, USA) staining as described in our previous report [9]. To evaluate brain edema, the water content was calculated in the brains, isolated 24 h after the operation, from five rats from each group according to the same procedure as used in our previous study [10].
Serum glucose determination. Blood samples were allowed to clot for 1 h at room temperature, after which time serum samples were collected by centrifugation at 12,000g for 10 min. Glucose concentration was determined using the Glucose Diagnostics Kit (Sigma) according to the manufacturer’s suggested protocol.
