Immunostaining showed that COX-2 was occasionally expressed in liver of WT mice. In contrast, strong COX-2 protein (+)-MK 801 was demonstrated in hepatocytes in TG mice (Fig. 1A1). Accordingly, PGE2 level in liver of TG mice was significantly higher than that of WT mice, indicating an elevated COX-2 activity in TG mice (Fig. 1A2).
Fig. 1.
(A) COX-2 protein expression and activity in the liver of transgenic (TG) and wild type (WT) mice. (A1) Profound COX-2 expression in the liver was confirmed in TG mice by immunohistochemistry. COX-2 was expressed in very low level in the liver of WT mice. COX-2 protein mainly localized in cytoplasm of hepatocytes radiating out from central veins was observed in TG mice. (A2) COX-2 activity as demonstrated by production of PGE2 was 27-fold higher in TG mice compared with the WT mice. Data represent means ± SD with n = 6–10/group, ?P
