Effect of Lu on glycolytic flux and glucose uptake in tumor cells
The glycolytic flux of tumor MCOPPB trihydrochloride was examined by lactate production. As shown in Fig. 2B, compared with under normoxia, both 4T1 and MCF-7 cells produced significantly more lactate under hypoxia in spite of ATP production decreased. Under normoxia, Lu at 50 or 100 μM had no effect on the lactate and ATP yield in both tumor cells (date not shown), whereas it significantly decreased lactate and ATP production under hypoxia (Fig. 2B). Lu at 50 or 100 μM did not change the intracellular glucose and glucose uptake under hypoxia (Fig. 2C and D).
Effect of Lu on level of P-gp in tumor cells
P-gp which could export anti-cancer drugs such as Dox out of the cells decreases intracellular drug concentrations and confers drug resistance. We examined whether hypoxia-induced drug resistance was relation to P-gp over-expression. As shown in Fig. 3A, the P-gp expression in tumor cells had no difference between under normoxia and under hypoxia, and Lu at 50 or 100 μM had also no effect on level of P-glycoprotein under hypoxia.
