Inhibition of cell proliferation by siRNA-mediated knockdown of raptor. (A) SW1116 GTP-Binding Protein Fragment were transiently transfected with raptor siRNA or control siRNA for 48 h and the effects on cell proliferation were determined by MTT assay. (B) Knockdown of raptor reduced the colony-forming ability of SW1116. Cells were transfected with raptor siRNA or control siRNA for 24 h and then incubated in fresh medium for another 6 days. ?P cell cycle or pro-apoptotic factors [19], [20], [21], [22] and [23]. Moreover, proteasome inhibitor induces cell cycle arrest through the activation of bone morphogenetic protein signaling [24] and [25]. Proteasome inhibitor has been shown to down-regulate telomerase activity and survivin expression [26]. Here we demonstrate for the first time that proteasome inhibitor represses mTORC1 signaling and protein translation, whose inhibition is sufficient to lower proliferation in colon cancer cells.
