FBS-concentration-dependent expression of the DC maturation markers in SF-DCs. After 8 days of culture, the medium was changed to SF-HY containing varying concentrations of FBS and cultured for an additional 2 days. SF-DCs were stained with a panel of FITC-labeled Abs, PE-labeled anti-CD11c Abs and PI, to analyze the cell-surface markers. The gray histograms show the expressions on the unstained DCs, and the thin lines show the expressions of each maturation marker on the DCs. The percentage of positive TH 302 (M1) is shown in each histogram.

Time-dependent changes of the DC maturation marker expressions in FBS-DCs and SF-DCs. FBS-DCs were harvested at 8 days of culture with conventional medium. The harvested FBS-DCs were separated into two groups and one was continued with conventional medium and another was changed to HY medium without FBS (See Materials and methods). FBS-DCs and SF-DCs were harvested after 0, 1, and 2 days additional culture and stained with a panel of FITC-labeled Abs, PE-labeled anti-CD11c Abs and PI, to analyze the cell-surface markers. The gray histograms show the expressions on the unstained DCs, and the thin lines show the expressions of each maturation marker on the DCs. The percentage of positive cells (M1) is shown in each histogram.
