Knockdown of EGLN1, 2, 3 and FIH by siRNA molecules in Hep3B cells. Hep3B HRE luciferase BB94 were transfected with siRNAs for EGLN1, EGLN2, EGLN3, and FIH and the expression level of each gene was analyzed by real time qPCR 3 days after transfection. Mock transfected Hep3B HRE luciferase cell samples were used as control. E1.3 refers to the number 1 siRNA in the Dharmacon siRNA mix for EGLN1, E3.4 the number 4 siRNA for EGLN3, etc. Triplicate experiments were performed and the average and error bars plotted.

Effect of EGLN/FIH knockdown on VEGF and Epo mRNA levels. (A) Effect of EGLN/FIH knockdown on VEGF mRNA level. Hep3B HRE luciferase cell line was transfected with single or combinations of siRNAs for each HIF hydroxylase gene and the level of VEGF mRNA was analyzed 3 days post-transfection using real time qPCR. Mock transfected Hep3B HRE luciferase cell samples were used as control. (B) EPO and VEGF stimulation by HIF hydroxylase knockdown in comparison with hypoxia treatment. Hep3B cells were transfected with single or combinations of siRNAs for each HIF hydroxylase gene or treated with hypoxia overnight. The level of Epo and VEGF mRNA was analyzed 3 days post-transfection. Mock transfected Hep3B HRE luciferase cell samples were used as control. Triplicate experiments were performed and the average and error bars plotted.
