You have to keep the rings in the same hand for their effect to work.

Same hand. As in, all on one side or another of your inventory icon. Then yes they do work.

However, ring sets have been changed now and then (not recently) so make sure you have the right guide as to which rings are actually in each set.

If your set works, it will glow gold. You'll see it right away, quite obvious. The order of the rings withing your hand don't matter.
