I have seen a gay character done well exactly once, to my recollection. One of Scott Pilgrim's friend.
Be careful what you wish for. I get the feeling a gay protagonist, in today's media, would be a bit... campy.

Scott Pilgrim's friend was promiscuous, though. I wouldn't really call that a positive portrayal of a gay man.

I only saw the movie, and even so it was 3/5 done by the time I tuned in.

I have seen quite a few movies with gay main characters, but these are the only ones I can think off of the top of my head.

Also, the comic series "The Sandman" by Neil Gaiman has some gay characters. Most of them appear in the volume "A Game of You," but they aren't MAIN characters, however, they are focused on a lot.

In my opinion, if the main character's sexuality is not a focal point of the film, then there's absolutely no point in including that minor detail if its only purpose is to demonstrate the filmmakers are open minded and attempting to break society's boundaries. Also, why does a gay main character have to be a role model? Not all main characters are good people. If anything, perfectly positive gay main characters are unrealistic depictions.

With all it's cutscenes the Metal Gear series has a lot of gay characters in it including several of the main antagonists and the two of the Player Characters have an almost romantic chemistry with male supporting characters without it being a major part of their characters.

Also in Boondock Saints, the FBI agent is seen in bed with another guy, has no onscreen female love interest (in fact the only one with a female love interest is a supporting character's girlfriend who gets a breif scene and that's it.)

If they ever do a good gay main character, it will be something like Dumbledore from Harry Potter. It's a part of who the character is, but it doesn't define them.

Look at the persona game series. They do gay characters fairly well.

the new batwoman is a lesbian. there's also scandal savage in secret six. the earth two green lantern is also gay. but that's just comics.

there have been a lot more gay characters in the mainstream media though. there need to be more lesbians and trans* characters though and not just lesbian and trans* characters that are purely there for comedy or caricatures.

As for anime—
Revolutionary Girl Utena, Tokyo Godfathers, Strawberry Panic!,
Gravitation, Loveless, Sailor Moon, and Miyuki-chan in Wonderland—
to name a few, although I don't watch these so much these days.
Sadly, the lack of GLBT* characters in Western cartoons is shameful.

Either way, if you look closely, there have been GLBT* characters in series and films
for at least 30 years, it just isn't always obvious, and shouldn't have to be unless
the plot revolves around sexual or gender identity or sex in general.

Instead of wanting things to be one way, try looking at life a little differently,
because what you're looking for is already there, you just haven't been paying attention
or wanting characters who sexual identity are irrelevant to define them.

All I can think of is that one episode of Doctor Who
from the other year — Midnight — where the Powers That Be decided
that what that episode really needed was a lesbian. And, of course, there was
an obligatory 'O MAH GAWD, ITZ A DYKE' moment when they, out of nowhere,
brought up her sex life and she put the emphasis on 'she'. Yes, because
that was something that we wanted to know, needed to know,
and added to the character development, apparently.

I don't think this is a fair description of what happened. The Doctor has a talent for getting
people's trust and in the case of the episode "Midnight", he and Sky was talking about travelling,
which is why she mentioned being alone and he asked why, prompting the response she
gave that her partner wanted space. It was a way to show that, in the future,
GLBT* persons will be see as absolutely normal.

Granted, that's not the only example of GLBT* in Doctor Who. There are also characters—
Captain Jack Harkness (Pansexual), Ianto Jones (Bisexual), Lady Cassandra (Trans*),
and Madam Vestra & Jenny (lesbians & partners)—who are just a small sample of
GLBT* inclusion in the new production of this series.
