If you have the master file, you'll be able to do it more easily than anyone else can. As long as the lines and color are not on the background layer, all you have to do is delete the background and save the file as a .png. However, you will probably have to go in and replace the white areas in the drawing, assuming you relied on the white background to provide that filling.
